Abstract

Cibacron Blue, a monochlorotriazine anthraquinone dye was covalently coupled to Eupergit C, an acrylic polymer support containing oxirane groups. The developed affinity adsorbent matrix was effective in the purification of phospholipase A2 from soybean (Glycine max) seeds. The coupling chemistry applied in the obtention of the affinity system was simple, not altering the interaction between phospholipase A2 and the immobilized ligand. The system has the advantage that both, the ligand and the support are chemical and microbiologically stable. Performing a classical column chromatography approach, phospholipase A2 was purified about 23 folds (purification factor). In the experimental conditions, the enzyme recovery (activity units) was 13.4%. The purified enzyme obtained by this approach could be applied in biotechnological processes. Lysophospholipids, derivatives obtained in the hydrolysis reaction promoted by phospholipase A2, are potent biosurfactants useful in food industry and for medical purposes.
